The stage directions maybe? Such as
Sheila (gaily) What's this about streets?
Here, "gaily" suggests she said it in a cheerful, light-hearted manner.
When reading out loud, emphasise the italics (slanted writing like this).
I didn't do it! - here you would emphasise the "didn't".
