A substance which contains unstable atomic nuclei is considered to be radioactive. Radioactive decay is a random process that occurs at the level of individual atoms.
Nicks are the gaps present in between the DNA strands where some nucleotide are missing. These gaps can be repaired by addition of dNTPs and DNA polymerase. One of the nucleotides is radiolabelled. The DNA polymerase has exonuclease activity and therefore, can fill up the gaps as well as replace the older DNA nucleotides of that strands with new nucleotides.
The resulting DNA would have radiolabelled DNA strand which can be isolated and used as a probe.
