A DNA polymerase I take out the RNA primer and then use DNA to fill in the spaces.
The gaps are filled up with DNA once the RNA primer is removed by DNA polymerase I. The procedure to eliminate the nicks, however, cannot be catalysed by DNA polymerase I. By creating the phosphodiester bond, DNA ligase—another enzyme—seals the nicks, creating a continuous sugar-phosphate backbone for the lagging strand. The Okazaki fragment is synthesised by DNA polymerase I enzyme in prokaryotes up until the preceding RNA primer. The enzyme then performs a simultaneous 5′–3′ exonuclease reaction, eliminating primer ribonucleotides up front and introducing deoxyribonucleotides in their place.
